Glucose Ketone Index May Offer a Broader Measure of Metabolic Health, Suggests Review

USA: A review published in Frontiers in Science suggests that the glucose ketone index (GKI)—a finger-prick measurement of the ratio between blood glucose and ketone levels—could help monitor metabolic health, particularly in people with cancer and chronic diseases. Researchers suggest that GKI may provide a more comprehensive picture of whole-body metabolic and endocrine function than blood glucose or body weight alone, potentially making it a useful biomarker for clinical monitoring. 

The review, authored by Derek C. Lee from the Biology Department at Boston College, United States, and colleagues, highlights the growing burden of chronic noncommunicable diseases (NCDs), which now account for the majority of disability-adjusted life years worldwide. The authors note that cancer is among the leading causes of death and may soon surpass heart disease as the leading cause of mortality in the United States. They argue that improving metabolic health is central to preventing and managing many of these conditions.
The researchers noted that mitochondrial dysfunction, marked by impaired cellular energy production, is linked to several chronic conditions, including cancer, cardiovascular disease, type 2 diabetes, obesity, and neurodegenerative disorders. Poor diet, physical inactivity, and environmental toxins may contribute to this dysfunction.
The review states that the glucose ketone index (GKI), originally developed to monitor adherence to ketogenic therapy in cancer patients, is calculated using the ratio of blood glucose to the ketone body β-hydroxybutyrate (βHB). The authors suggest that GKI may provide a more comprehensive measure of metabolic health and chronic disease risk than blood glucose alone.

However, they emphasize that further clinical studies are needed to validate its broader role in preventing and managing cancer and other chronic diseases.
