Alzheimer's Research Uncovers a Potentially Overlooked Contributor to the Disease - Video

Could scientists have overlooked another protein responsible for damaging brain cells in Alzheimer's disease? A new scientific commentary suggests that a little-studied peptide called P3, previously considered harmless, may contribute to Alzheimer's disease by forming potentially toxic protein deposits in the brain.

Published in the journal ChemBioChem, the commentary by researchers at the University of California, Santa Cruz, highlights emerging evidence challenging longstanding assumptions about the biological role of P3 in Alzheimer's disease.

For decades, Alzheimer's research has largely focused on amyloid beta (Aβ), a peptide that accumulates in the brain and forms protein aggregates associated with neuronal damage and cognitive decline.

However, despite extensive research and numerous clinical trials targeting amyloid beta, treatment outcomes have remained limited, prompting scientists to investigate additional disease mechanisms.

The commentary highlights evidence that P3, a smaller peptide derived from amyloid precursor protein (APP), can also form amyloid aggregates and fibrils.

Unlike amyloid beta, which is produced through the action of beta-secretase and gamma-secretase enzymes, P3 forms when APP is processed by alpha-secretase followed by gamma-secretase.

Experimental studies conducted over the past five years have demonstrated that P3 can form protein aggregates even faster than amyloid beta, contradicting earlier assumptions that it remains soluble and biologically harmless.

Researchers also identified evidence suggesting that P3 may be toxic to neurons, although its observed toxicity appears lower than that of amyloid beta.

Importantly, emerging findings indicate that P3 may interact with amyloid beta, potentially influencing protein accumulation and neurotoxicity.

However, researchers emphasised that these findings are primarily based on experimental investigations, and P3's direct contribution to Alzheimer's disease in humans remains uncertain.

Further research could clarify the role of P3 in neurodegeneration and potentially identify new molecular targets for developing more effective Alzheimer's treatments.
